The Basic Principle of Soxhlet Extraction

Soxhlet extraction, also known as continuous extraction and Soxhlet extraction, is a method of extracting compounds from solid substances. Soxhlet extraction is used to determine crude fat content. Fat is widely present in the seeds and fruits of many plants. Determining the fat content can be used as an indicator to identify the quality of fat. Extraction methods are widely used at home and abroad, among which Soxhlet extraction is a recognized classic method and the preferred standard method for grain and oil analysis in my country. This method takes a long time, and fat extractors (Soxhlet extractors) are often used for extraction in laboratories.

Principle of “similar similarity.”

basic principle of soxhlet extractionIn the laboratory, a fat extractor (Soxhlet extractor) is used for extraction. The fat extractor uses the solvent reflux and siphon principle to continuously extract the solid matter by pure solvent, which saves the solvent extraction efficiency and high the efficiency. The solid material is ground before extraction to increase the area of solid-liquid contact. The solid material is then placed in a filter paper holder and placed in an extractor. The bottom end of the extractor is connected to a round-bottom flask containing a solvent, and is connected to a reflux condenser. The bottom flask is heated to boil the solvent, the vapor rises through the branch pipe of the extractor, is condensed and drops into the extractor, and the solvent is contacted with the solid for extraction. When the solvent surface exceeds the highest point of the siphon, the solvent containing the extract is siphoned back. The flask, thus extracting a portion of the material, is repeated such that the solid material is continuously taken as a pure solvent, and the extracted material is concentrated in the flask.

The core principle of Soxhlet extraction is to achieve efficient and continuous extraction of target compounds in solid samples through the circulation mechanism of solvent reflux and siphon action. The following is a detailed analysis of its working principle: ‌

1. ‌Solvent reflux process‌

‌Heating and vaporization‌: Add low-boiling point organic solvents (such as petroleum ether, ether) to the extraction bottle (flask), and the solvent vaporizes after heating, and the steam rises to the condenser through the connecting tube.
‌Condensation and dripping‌: The steam is cooled and liquefied in the condenser to form pure solvent droplets, which drip into the extraction tube containing the sample. The sample is usually ground and wrapped in a filter paper tube to increase the contact area.
‌Extraction target‌: The solvent contacts the sample in the extraction tube, dissolves the lipids or target compounds (such as fatty acids) therein, and forms a mixed solution containing the extract.

2. ‌Siphon and circulation mechanism‌

‌Liquid level triggers siphon‌: When the solvent level in the extraction tube reaches the top of the siphon tube, the mixed solution is completely refluxed back into the extraction bottle due to the siphon effect, completing an extraction cycle.
‌Repeat extraction‌: The solvent in the extraction bottle is heated and vaporized again, and the above process is repeated. ‌Fresh, pure solvent is used in each cycle to ensure efficient extraction until the target compound is completely extracted (usually taking 6-12 hours).
